چکیده
Abstract Pediatric low-grade glioma incidence has been rising in the U.S. over last 20 years, concurrent with a rate of obesity both adult and pediatric populations. Recently, children obese mothers have demonstrated to increased rates several tumors, including brain tumors. Importantly, is driven large part by diet, given abundance accessibility high-fat, high-sugar food choices. High-fat diet exposure previously promote proliferation glial differentiation embryonic neuroglial progenitor cells (NPCs) around third ventricular zone (TVZ), suggesting that utero an obesogenic might affect formation tumors derived from these cells, such as Neurofibromatosis Type 1 (NF1)-related optic pathway (OPG). Based on data, we hypothesized maternal would increase Nf1-OPG through intrinsic effects tumor cell origin. We progeny dams exposed HFHS during preceding gestation WT Nf1-heterozygous TVZ NPCs vivo. Progeny non-obese this similar phenotype, were related rather than weight. then assessed how affected formation. determined increases penetrance two low-penetrance models NF1-OPG. Finally, resulted earlier onset high-penetrance model. Taken together, findings suggest formation, origin while still utero. It may also implications for clinical prognosis, age negatively associated visual outcomes

The cell of origin dictates the temporal course of neurofibromatosis-1 (Nf1) low-grade glioma formation
Low-grade gliomas are one of the most common brain tumors in children, where they frequently form within the optic pathway (optic pathway gliomas; OPGs). Since many OPGs occur in the context of the Neurofibromatosis Type 1 (NF1) cancer predisposition syndrome, we have previously employed Nf1 genetically-engineered mouse (GEM) strains to study the pathogenesis of these low-grade glial neoplasms....
